RIPA buffer contains the nonionic detergent, Nonidet® P-40 Substitute, plus two ionic detergents, sodium deoxycholate and SDS. Simply, resuspend cells in cold RIPA Lysis Buffer. Incubate on ice, and then pass the cells through a pipette several times. Pellet the cellular debris and transfer the cell lysate to a fresh microcentrifuge tube.
